Design Notes

When anti-CRISPR (acr) promoter inhibitor Anti-CRISPR-associated 1 (Aca1) is mutated to Aca1R44A, Aca1R44A can no longer bind to the promoter region, therefore the acr promoter will not be inhibited. When green fluorescent protein is inserted after the acr promoter, the efficiency of the promoter can be measured.

Through the experiments we did, we found Navitoclax can inhibit Aca1 effectively and thus increase genes downstream Acr promoter significantly.
